脱盐时, G 25,G50如何选择?
G25和G50的区别是孔径有明显差异。G50的孔径较大。更适宜处理分子量较大的目标蛋白,如抗体等等。
据此建议:当目标蛋白的分子量大于1500kda时应当用G50。其他情况用G25。根据我们的测试。总体来说,G25的脱盐效果更高一些。因此,在对脱盐效果要求较高时尽量使用G25。用G25处理大分子量蛋白时可能会导致流速低,操作时间拉长。这种情况对于脱盐效果有一定的影响。但通常还是可以获得理想的结果的。

根据拟处理的样品体积按照上样量不超过胶床体积30%选择合适的预装柱。例如:如果拟脱盐的目标蛋白体积为3ml,应当选胶床体积为9m左右l的脱盐柱。 此外,脱盐柱的胶体积应当有一定的高度,胶床高度过短的柱子脱盐效果不好。据本公司测定,胶床高度为2厘米时,一次脱盐率约为75%。8~12厘米高度一次脱盐可可获得99%或更高的脱盐率。 |
